Overview

Leucine-rich repeat-containing G protein-coupled receptor 6 (LGR6) is a member of the G protein-coupled receptor superfamily distinguished by 17 leucine-rich repeats flanked by cysteine-rich motifs in its large N-terminal extracellular domain. LGR6 serves as a high-affinity receptor for R-spondins 1–3, enhancing canonical Wnt/β-catenin signaling primarily through increased LRP6 phosphorylation. It is not coupled to canonical heterotrimeric G-protein signaling upon ligand binding. LGR6 identifies populations of multipotent stem cells in tissues such as skin and is involved in tissue regeneration and tumor suppression, with roles described in several cancers, and modulates cardiac hypertrophy via metabolic pathways[1][2][4][5][6][8].
