Overview

Leucine-rich repeat-containing protein 1 (LRRC1), also known as LANO, is a member of the LAP (leucine-rich and PDZ domain-containing) family that plays an essential role in maintaining epithelial cell polarity and organization[2][4]. This 524-amino-acid protein is structurally defined by its leucine-rich repeat motifs, which facilitate protein-protein interactions crucial for cellular architecture and signal transduction[1][3]. LRRC1 is expressed highly in polarized cell types during development and is implicated in tissue morphogenesis[4]. In cancer biology, LRRC1 is overexpressed in hepatocellular carcinoma (HCC) and some other tumor types, correlating with poor prognosis and aggressive disease features[2]. Mechanistically, it promotes cell proliferation, invasion, and migration, likely by modulating cytoskeletal organization and metabolic pathways (including amino acid and carbohydrate metabolism)[2][4]. LRRC1 integrates into complex networks regulating cell polarity, and its mislocalization or overexpression may disrupt tissue architecture, a key event in malignancy development[2][4]. At present, LRRC1 is viewed as a potential prognostic marker and emerging therapeutic target rather than a direct target for approved drugs, with ongoing research into its detailed molecular functions and therapeutic applications.
