Leucine-rich repeat-containing protein 14 (LRRC14) is a member of the LRR protein family, characterized by multiple leucine-rich repeats that create a protein–protein interaction scaffold[1][3]. LRRC14 acts as a negative regulator of Toll-like receptor (TLR)-mediated signaling, suppressing downstream NF-κB activation by binding to IKKβ and disrupting its interaction with NEMO, thereby interfering with the phosphorylation and activation of NF-κB[2][3]. Its expression can be modulated by inflammatory stimuli, and although its full biological role is not completely understood, studies implicate it in modulating inflammation and possibly immune homeostasis[2][3]. LRRC14 is not a classical therapeutic target such as a receptor, enzyme, or transporter, and there are currently no known drugs that directly target it[2][3]. It has been linked in genetic studies to Rapadilino syndrome and retroperitoneum carcinoma, but no established clinical biomarker or therapeutic application exists for LRRC14[3].
