Overview

Leucine-rich repeat-containing protein 26 (LRRC26) is a single-transmembrane auxiliary protein and the γ1 subunit of large-conductance, calcium- and voltage-activated potassium (BK) channels. LRRC26 is required for converting BK channels from high-voltage to low-voltage activation, enabling BK current under physiological conditions, especially in secretory epithelial cells and arterial smooth muscle. It modulates potassium flux, membrane potential, and secretory functions, and is crucial for proper vasodilation and regulation of glandular secretion. LRRC26 also influences normal myoblast differentiation and is linked to the malignant progression of triple-negative breast cancer, likely through both potassium channel-dependent and independent mechanisms. Its gene is conserved across many species, with predominant expression in secretory glands and some smooth muscle tissues[1][2][3][4][6].

Mechanism of action

Alters voltage and calcium sensitivity of BK (large-conductance, voltage and calcium-activated potassium) channels to enable channel opening at physiological voltages and low calcium. Drugs that modulate BK channel activity (inhibitors or activators) may indirectly affect or require LRRC26 function for their effects.
