Leucine-rich repeat-containing protein 3B (LRRC3B) is a membrane-associated protein characterized by leucine-rich repeat domains and is encoded by the LRRC3B gene, commonly located at chromosome 3p24.1. It acts primarily as a tumor suppressor and is frequently silenced by promoter hypermethylation in many epithelial cancers, including gastric, lung, renal, breast, ovarian, prostate, and colorectal cancers. Reduced LRRC3B expression is associated with poor prognoses and advanced tumor stage. LRRC3B regulates multiple cellular processes, including cell proliferation, cell cycle, apoptosis, cell adhesion, signal transduction, and plays a role in immune surveillance by influencing immune cell infiltration in the tumor microenvironment. DNA methylation of the LRRC3B promoter serves as a promising biomarker for cancer risk assessment, early detection, prognosis, and response prediction to cancer immunotherapy. No approved drugs are known to directly target LRRC3B; its clinical significance is mainly as a tumor suppressor and prognostic biomarker.
