Overview

Leucine-rich repeat-containing protein 42 (LRRC42) is a small nuclear protein characterized by the presence of two leucine-rich repeat motifs, which are structural motifs facilitating protein-protein interactions[4][2]. In normal human tissues, LRRC42 expression is highly testis-specific, but it is significantly upregulated in most lung cancers[4][6][2]. Experimental data show that knockdown of LRRC42 reduces lung cancer cell growth, while its overexpression promotes proliferation, suggesting an oncogenic role in this context[4][2][6]. LRRC42 interacts with proteins such as GATAD2B (a component of the MeCP1 complex) and MBD3, participating in the repression of transcription for targets such as the cyclin-dependent kinase inhibitor p21Waf1/Cip1—an established cell cycle regulator and tumor suppressor[2][4][6]. The LRRC42 protein has not been reported to interact with any approved drugs or to be the direct target of therapeutic agents to date. Its biological roles highlight the importance of LRR domain-containing proteins in mediating critical processes such as gene regulation and cell proliferation, especially in the context of carcinogenesis[2][4][6][1].
