Overview

Leucine-rich repeat-containing protein 52 (LRRC52) is a single-pass transmembrane auxiliary subunit belonging to the leucine-rich repeat family, primarily expressed in human testis and sperm. LRRC52 closely associates with Slo3 and BK potassium channels, dramatically shifting their gating properties and making them and their activity highly sensitive to pH and voltage changes. In sperm, LRRC52 is required for potassium current activation (KSper), making it essential for male fertility. It also regulates channel localization and stability in certain excitable cells. LRRC52's role is primarily functional modulation; there are currently no drugs directly targeting LRRC52, and loss of function is tied to infertility rather than any broader disease phenotype[1][2][3][5][7].
