Leucine-rich repeat-containing protein 55 (LRRC55)

Target
LRRC55
Molecular classification
Ion channel auxiliary subunit, Single transmembrane protein, Leucine-rich repeat (LRR)-containing membrane protein
01

Overview

Leucine-rich repeat-containing protein 55 (LRRC55) is a single-pass transmembrane protein and a member of the leucine-rich repeat (LRR) family. It acts as an auxiliary gamma (γ3) subunit of the BK (large-conductance, voltage- and calcium-activated potassium) channel, where it distinctly modulates channel gating by producing a negative shift in the voltage dependence of activation. LRRC55 is mainly expressed in the brain and is structurally similar to other BK channel gamma subunits (LRRC26, LRRC38, LRRC52). It plays a role in the physiological regulation of neuronal excitability and cell signaling by influencing the properties of the BK channel complex, though direct implications in disease or as a drug target are not yet firmly established[1][2][3][4][6].

02

Mechanism of action

Modulates voltage dependence and gating of the BK (large conductance, calcium- and voltage-activated potassium) channel by shifting activation threshold toward more negative potentials; this effect occurs independently of cytosolic calcium[1][2][3][6].

03

Biological functions

Regulation of ion channel gating (specifically BK channel)Modulation of neuronal and non-neuronal electrical signaling
04

Disease associations

Other (differential tissue expression and potential roles in neurological functions; specific disease associations not clearly established)
05

Safety considerations

None directly reported; as an auxiliary subunit, off-target effects would be linked to broader disruption of BK channel function in tissues such as the brain[1][2].
06

Interacting drugs

None directly reported for LRRC55; interacts functionally with the pore-forming BK channel, which is a target for some potassium channel modulators[1][3][6].
07

Biomarkers

None reported specifically for LRRC55[3].
