Leucine-rich repeat-containing protein 59 (LRRC59) is a membrane protein localized to the endoplasmic reticulum and is notable for facilitating nuclear import of fibroblast growth factor 1 (FGF1), but not FGF2. It also possesses cadherin and RNA binding activity, and is involved in intracellular signal transduction in human cells. LRRC59 is overexpressed in bladder cancer, where its increased expression correlates with poorer prognosis and aggressive tumor behavior; it serves as a novel prognostic biomarker for this disease. Knockdown of LRRC59 leads to reduced proliferation and migration of bladder cancer cells and affects epithelial-mesenchymal transition markers, implicating its role in cancer pathobiology. As a member of the large family of leucine-rich repeat (LRR) proteins, it may also share general features of protein–protein interaction scaffolding and immune-related functions seen in other family members. However, its direct involvement in drug interactions, specific mechanisms of action for targeted therapies, and safety profiles have not been established in the literature available.
