Overview

Leucine-rich repeat-containing protein 7 (LRRC7), also known as Densin-180 or LAP1, is a brain-specific scaffolding protein found predominantly in the postsynaptic density (PSD) of neurons[1][3][5][9]. It contains 16 leucine-rich repeats at the N-terminus and a PDZ domain at the C-terminus, and it is required for normal synaptic spine architecture and function[1][9]. LRRC7 interacts with various synaptic proteins, including CaMKII-alpha, actin-binding proteins, NMDA receptor complexes, and Shank[1]. It is necessary for proper localization of proteins such as DISC1 and metabotropic glutamate receptor 5 to the PSD and influences glutamatergic synaptic plasticity[3][9]. Variants in LRRC7 have been implicated in neurodevelopmental disorders, including autism spectrum disorders, cocaine dependence, developmental delay, and potentially epilepsy, although definitive disease causality is still under evaluation[3][5]. LRRC7 is not classified as a classic therapeutic target such as a receptor or enzyme, and there are no known drugs directly targeting this protein.
