Leucine-rich repeat-containing protein 74A (LRRC74A) is a cytoplasmic protein encoded by the LRRC74A gene on chromosome 14q24.3. It is comprised of 488 amino acids, with a predicted molecular weight of ~55 kDa and an isoelectric point of 5.22. The protein features eight leucine-rich repeat domains, a hallmark structure supporting protein-protein interactions, and has structural similarity to ribonuclease inhibitor proteins. LRRC74A is most highly expressed in the testis, salivary gland, and pancreas, though at generally low levels. Mutations in LRRC74A have been associated with increased mortality in COVID-19 and associations with twin-to-twin transfusion syndrome and primary ciliary dyskinesia. Although its precise biological role is not fully characterized, its structural features suggest a function in intracellular protein interactions; in some model organisms, it is predicted to bind calcium ions. There are currently no known drugs or targeted therapies acting on LRRC74A, and it is not classified as a therapeutic target.
