Leucine-rich repeat neuronal protein 2 (LRRN2) is a single-pass transmembrane protein that belongs to the leucine-rich repeat superfamily, predicted to participate in cell adhesion and signal transduction and to function as a signaling receptor[1][2][3][4][6]. It plays a role in vertebrate nervous system development, notably in the guidance of motor axons during embryogenesis, where it acts downstream of regulators like Hoxb1 and Sonic hedgehog in chick neural development[1]. LRRN2 is also found to be amplified and overexpressed in malignant gliomas, suggesting a role in oncogenesis and making it a candidate gene in cancer-related chromosomal amplicons[2][3]. It is associated with other diseases, including autism spectrum disorder and ependymoma, though its precise molecular and pharmacological functions remain incompletely characterized[3]. The encoded protein shares homology with cell-adhesion molecules and signal-transducing receptors, but no approved drugs or known pharmacological modulators are currently described for this target[3][4].
