Overview

Leucine-rich repeat neuronal protein 4 (LRRN4) is a type I transmembrane protein of the leucine-rich repeat (LRR) family, structurally characterized by multiple extracellular LRR motifs and a fibronectin type III (FN-III) domain[1][5]. It is expressed in the nervous system (e.g., hippocampus, dorsal root ganglia), heart (particularly ventricular cardiomyocytes), lung, and ovary[1][5]. In the heart, LRRN4 is implicated in maintaining normal cardiac morphology and function and is downregulated in dilated cardiomyopathy but not ischemic heart disease, positioning it as a potential therapeutic target for specific forms of cardiomyopathy[1]. In the nervous system, LRRN4 is required for hippocampus-dependent memory consolidation and may act as a synaptic adhesion molecule[1][4]. In cancer, especially colorectal cancer, LRRN4 is upregulated, promoting cell proliferation, DNA synthesis, and inhibiting apoptosis—functions apparently mediated via the Ras/MAPK and PI3K pathways[2]. Diseases associated with LRRN4 include tracheal cancer, colorectal cancer, and arrhythmogenic right ventricular cardiomyopathy[3]. There are no known drug interactions or therapeutics specifically targeting LRRN4 as of this time.
