Overview

Leucine-rich repeats and immunoglobulin-like domains protein 3 (LRIG3) is a member of the LRIG family of single-pass transmembrane proteins characterized by ectodomains containing several leucine-rich repeats and immunoglobulin-like domains. LRIG3 modulates the activity of multiple receptor tyrosine kinases (including EGFR, MET, and PDGFRα) primarily in cell signaling pathways involved in cell proliferation, migration, and differentiation. Functionally, LRIG3 is implicated in both positive and negative regulation of growth factor receptor signaling, with context-dependent effects in different tissues and cancer types. Knockout studies in mice show LRIG3 is essential for normal neural crest, craniofacial, cardiac, and inner ear development. In cancer, LRIG3 is reported to modulate malignant cell behavior and may act as either a tumor suppressor or promoter. No drugs currently target LRIG3 clinically, though its expression and release as a soluble ectodomain (sLRIG3) present possible future biomarker or therapeutic applications[1][2][3].
