Overview

Leucine zipper and EF-hand containing transmembrane protein 2 (LETM2) is a mitochondrial inner membrane protein, structurally characterized by a conserved LETM1 domain, a transmembrane segment, a leucine zipper motif, and a C-terminal EF-hand domain[2][5]. In mammals, LETM2 is predicted to function as a transmembrane transporter involved in mitochondrial calcium ion transmembrane transport and mitochondrial organization[2][4][5]. LETM2 shares homology with LETM1, but is less studied; mammalian LETM2 is noted for its restricted expression in testis and sperm, suggesting a specialized physiological role, possibly in spermatogenesis[3]. In plants (Arabidopsis), LETM2 orthologs are essential for efficient mitochondrial protein translation and for normal mitochondrial function and development[1]. There are no known clinically relevant small-molecule drugs or established disease associations for human LETM2, and its role as a therapeutic target has not been established[2][5].
