Leucine zipper tumor suppressor 1 (LZTS1) is a ubiquitously expressed tumor suppressor protein containing a leucine zipper motif, often deleted or downregulated in various malignancies. LZTS1 regulates cell-cycle progression by interacting with key mitotic regulators such as Cdk1/cyclinB1 and CDC25C phosphatase. Its loss or inactivation leads to uncontrolled cell proliferation, impaired mitotic control, and heightened metastatic potential in tumors. LZTS1 also influences epithelial cell differentiation and E-cadherin-mediated adhesion; its downregulation is linked to promotion of epithelial-mesenchymal transition (EMT) and poor prognosis, especially in gastric and colorectal cancers. LZTS1 is considered a prognostic biomarker but is not currently a direct molecular drug target.
Inhibition of cell proliferation via interaction with Cdk1/cyclinB1 complex and CDC25C phosphatase. Suppression of mitosis, leading to decreased tumor growth. Stabilization of E-cadherin-mediated adherens junctions, impacting cell adhesion and differentiation. Negative regulation of the PI3K-AKT pathway in some contexts.
