Leukemia Inhibitory Factor Receptor (LIFR), also known as CD118, is a cell surface receptor that mediates the actions of leukemia inhibitory factor (LIF) and other cytokines in the interleukin-6 family. Upon ligand binding and complex formation with gp130, several intracellular cascades are activated, including JAK/STAT, MAPK, and PI3K pathways. LIF/LIFR signaling has pleiotropic effects depending on tissue context, influencing cell proliferation, differentiation, survival, bone metabolism, neural development, embryogenesis, placental formation, maternal receptivity to blastocyst implantation, immunity, and tissue regeneration. A soluble form of the receptor exists which can inhibit biological activity by sequestering free ligand.
Agonist: LIF binding induces heterodimerization with gp130, activating JAK/STAT, MAPK, and PI3K signaling pathways. Antagonist: Soluble LIFR sequesters LIF, preventing it from binding to membrane-bound LIFR.
