The **Leukemia inhibitory factor receptor–gp130 complex** is a heterodimeric cell-surface receptor complex formed by the binding of the cytokine leukemia inhibitory factor (LIF) to the LIF receptor beta (LIFRβ), which then associates with the signal-transducing co-receptor glycoprotein 130 (gp130, also called IL6ST)[2][5][6][9]. Both LIFR and gp130 are class I cytokine receptors. Upon LIF binding, the receptor complex activates several downstream intracellular signaling cascades—most notably the JAK/STAT pathway (primarily STAT3), as well as MAPK/ERK and PI3K/AKT pathways—thereby regulating a broad range of cellular processes including survival, proliferation, differentiation, and apoptosis[2][4][6][8]. The LIFR–gp130 complex is essential for normal development, immune function, and tissue regeneration, but is also implicated in multiple diseases such as cancer, inflammation, and neurodegeneration. Dysregulated signaling through this complex can promote tumor progression, metastasis, and therapeutic resistance, making the receptor complex a promising therapeutic target and prognostic marker in oncology and beyond[2][4][8].
