LAIR1 is an inhibitory immune receptor expressed on most peripheral blood mononuclear cells. It binds to collagens and collagen-domain-containing proteins, leading to the recruitment of SHP phosphatases and inhibition of immune cell activation. It plays a role in maintaining immune tolerance and is implicated in autoimmune diseases, cancer, and infectious diseases.
Upon ligand binding (collagens), ITIM motifs are phosphorylated, recruiting SHP family phosphatases (SHP-1/SHP-2) to inhibit cellular activation pathways.
