Leukocyte-endothelial interaction refers to the series of molecular and cellular events that mediate the recruitment of leukocytes (white blood cells) from the bloodstream to sites of inflammation, infection, or tissue injury. This process is a hallmark of the inflammatory response and involves a tightly regulated multistep cascade between circulating leukocytes and vascular endothelial cells. The interaction occurs through several sequential steps: Tethering/Rolling, Activation, Firm Adhesion, and Transmigration. Key molecules involved include selectins, integrins (particularly β2-integrins), ICAMs/VCAMs, and PECAM-1. Dysregulation or excessive activation contributes to tissue damage seen in inflammatory diseases. Therapeutic strategies have targeted various steps to reduce inappropriate inflammation.
