Leukocyte immunoglobulin-like receptor subfamily A member 1 (LILRA1) is a human type I transmembrane glycoprotein belonging to the immunoglobulin superfamily. It is primarily expressed on myeloid cells, such as monocytes, macrophages, dendritic cells, and to some extent on B cells and subsets of T cells and NK cells[1][2][3]. LILRA1 contains extracellular immunoglobulin-like domains that allow it to function as an activating receptor, signaling through association with the FcεRIγ chain. Upon ligand binding—most notably certain HLA class I molecules—LILRA1 triggers activation pathways leading to immune cell proliferation, maturation, and survival, predominantly via downstream ITAM signaling and kinase activation[1][2]. LILRA1 and related receptors fine-tune immune responses by modulating cell activation thresholds, helping distinguish self from non-self and balancing activation and tolerance. While implicated in inflammation and infection, the specific disease associations for LILRA1 are less characterized compared to other LILR family members[1][2]. As of now, there are no drugs targeting LILRA1 directly in clinical use or advanced development, and its targeting potential is still being evaluated.
