Overview

Leukocyte immunoglobulin-like receptor subfamily A member 5 (LILRA5) is a transmembrane protein and a member of the immunoglobulin superfamily, classified as a Group 2 activating leukocyte immunoglobulin-like receptor. It is primarily expressed on monocytes and neutrophils. LILRA5 is structurally distinct from Group 1 LILRs, lacking affinity for classical major histocompatibility complex (MHC) class I molecules, and is presumed to bind non-MHC ligands to mediate activation signals. LILRA5 activation induces calcium flux and release of proinflammatory cytokines, implicating it in innate immune responses. Genetic variants in LILRA5 have been associated with increased risk of late-onset sporadic Alzheimer’s disease. Its crystal structure confirms substantial conformational and charge differences compared to related MHC-binding receptors. Four spliced transcript variants encode distinct isoforms of this protein.
