Leukocyte immunoglobulin-like receptor subfamily A member 6 (LILRA6) is a cell surface receptor of the immunoglobulin superfamily, predominantly expressed on myeloid and some lymphoid cells[5][1]. It is classified as an activating leukocyte immunoglobulin-like receptor (LILRA), with a structure characterized by extracellular immunoglobulin-like domains, a transmembrane region, and a short cytoplasmic tail that associates with the Fc receptor gamma chain (FcRγ) to mediate activating intracellular signals[1]. LILRA6 is involved in the recognition of MHC class I molecules and in the modulation of both innate and adaptive immune responses, particularly the activation and regulation of macrophages and cytokine secretion through pathways such as JAK-STAT, NF-κB, PI3K/AKT, and ERK1/2 MAPK signaling[1][5]. Genetic variation in LILRA6 has been associated with susceptibility to immune-mediated diseases, including atopic dermatitis and various inflammatory or infectious conditions[1]. No known approved therapeutic drugs selectively target LILRA6 as of now, and its potential as a biomarker or therapeutic target for immunomodulatory therapies remains an area of research interest[5][1].
