Overview

Leukocyte immunoglobulin-like receptor subfamily B member 4 (LILRB4), also known as ILT3, LIR-5, or CD85k, is an inhibitory immune receptor with significant roles in immune regulation and disease pathology. It is primarily expressed on monocytic cells and upregulated in certain pathological conditions like AML and solid tumors. LILRB4 acts as an immune checkpoint molecule, negatively regulating activation signals by recruiting SHP phosphatases upon ligand engagement and ITIM phosphorylation. Targeting LILRB4 may enhance anti-tumor immunity, making it a promising candidate for cancer immunotherapy.
