Overview

Leukocyte-specific transcript 1 protein is a membrane-bound transmembrane adaptor protein, predominantly expressed in leukocytes of the myeloid lineage, where it localizes to tetraspanin-enriched microdomains. It lacks enzymatic activity but contains conserved phosphotyrosine-based motifs (ITIM and ITIM-like) that allow it to interact with SHP-1 and SHP-2 phosphatases, thereby contributing to the negative regulation of immunological signaling. LST1 modulates immune responses and can inhibit lymphocyte proliferation. It can also induce morphological changes such as filopodia formation and may play a role in dendritic cell maturation. While associated with alternative splicing and multiple isoforms, only the LST1/A isoform is clearly conserved and detected in vivo. Genetic variation in LST1 has been linked with isolated cleft palate but lacks well-characterized associations with major common diseases[1][2][3][7].
