Leukotriene B₄ Receptor (LTB₄R)

Target
LTB₄R
Molecular classification
G protein-coupled receptor, Receptor
01

Overview

The leukotriene B₄ receptor (LTB₄R) is a G protein-coupled receptor that mediates the cellular effects of leukotriene B₄ (LTB₄), a potent lipid mediator derived from arachidonic acid. LTB₄R plays a central role in inflammation by regulating the recruitment and activation of immune cells, particularly leukocytes. There are two main subtypes: BLT1 (high-affinity) and BLT2 (lower-affinity). Antagonists targeting these receptors have shown efficacy in preclinical models for autoimmune diseases, arthritis, transplantation rejection prevention, and are being explored for metabolic/inflammatory disease treatment. Drug design faces challenges due to the broad ligand-binding pocket leading to specificity issues.

Mechanism of action

Antagonists bind to the LTB₄ receptor, preventing LTB₄ binding and subsequent G protein activation, thereby inhibiting downstream signaling pathways and reducing inflammation.
