Overview

**LIFR antisense RNA 1 (LIFR-AS1)** is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the LIFR gene locus on human chromosome 5p13.1[3][2]. It does not encode a protein[3]. LIFR-AS1 has been studied primarily for its roles in various cancer types, where its expression appears to influence cell proliferation, migration, invasion, and apoptosis, sometimes acting as a tumor suppressor and other times as an oncogenic factor, depending on the cancer context[1][2][4]. Mechanistically, LIFR-AS1 interacts with multiple microRNAs (such as miR-29a, miR-4698, miR-197-3p, miR-942-5p) as a competitive endogenous RNA (ceRNA), thereby regulating downstream targets including TNFAIP3 and MTUS1[1][7][2]. LIFR-AS1 also impacts signaling pathways relevant to cancer, such as inhibiting the MEK/ERK pathway and modulating the activity of cell cycle regulators like Cdc25B and Cdk1[2]. Reduced expression of LIFR-AS1 in various tumors is often associated with poor prognosis, increased tumor size, and higher metastatic potential, highlighting its potential as a biomarker for cancer diagnosis and prognosis[2][4][5]. There are currently no drugs known to directly target LIFR-AS1, and it is not considered a canonical therapeutic target such as a receptor or enzyme, but rather a regulatory RNA of emerging interest in oncology research[1][3][5].
