Overview

LCORL (Ligand-dependent nuclear receptor corepressor-like protein) is a DNA-binding transcription factor with critical roles in sperm development and regulation of skeletal growth and body size. Genetic variations in LCORL are robustly associated with height and skeletal morphology in humans, as well as rapid growth and body conformation in several animal species. LCORL may act in different isoforms—as a transcription factor and potentially as an accessory protein for polycomb repressive complex 2 (PRC2), which is involved in epigenetic silencing and developmental gene regulation. Despite significant research implicating LCORL in developmental biology and animal breeding, there is currently no evidence for direct pharmacological targeting, nor known small molecule or biologic drugs that interact with LCORL.
