Overview

LIM and calponin homology domains-containing protein 1 (LIMCH1) is a cytoskeleton-associated protein encoded by the *LIMCH1* gene in humans[1][3]. It contains an N-terminal calponin homology (CH) domain, a central coiled-coil region, and a C-terminal LIM domain, which confer actin and myosin-binding functions[2][4]. LIMCH1 localizes to contractile actin stress fibers, where it directly binds to the head of non-muscle myosin IIa (NM-IIA) but not NM-IIB, via its N-terminal region. LIMCH1 positively regulates non-muscle myosin IIa activity by promoting the phosphorylation of myosin regulatory light chain (MRLC/MYL9), which in turn enhances actin stress fiber assembly and stabilizes focal adhesions. Loss of LIMCH1 expression reduces actin stress fibers, decreases central focal adhesion maturation, and enhances cell motility. Although it modulates major processes in cell shape and migration and is up/downregulated in several cancer cell lines, there are currently no clinically established drugs targeting LIMCH1 nor established biomarkers or direct safety concerns associated with targeting this protein[1][2][4].
