Overview

LIM domain and actin-binding protein 1 (LIMA1) is a cytoskeleton-associated protein involved in the regulation and stabilization of actin filaments in eukaryotic cells[2][3]. It was originally identified as a tumor suppressor, frequently lost in various epithelial cancers, and is crucial for maintaining cell shape, cell adhesion, and membrane dynamics[2]. LIMA1 exists as two main isoforms (alpha and beta) and crosslinks actin filaments, inhibits filament depolymerization, and regulates cell motility and proliferation. It also plays a central role in cholesterol homeostasis by modulating intestinal cholesterol absorption through interaction with NPC1L1 and associated protein trafficking machinery. Downregulation of LIMA1 has been linked to enhanced tumor growth, invasion, and metastasis, while certain LIMA1 mutations are related to altered plasma cholesterol levels[1][2][3].
