Overview

LIM domain containing preferred translocation partner in lipoma (LPP) is a multifunctional LIM domain protein that localizes to focal adhesions and cell–cell contacts, shuttling between the cytoplasm and nucleus where it serves as a scaffold protein for structural and signaling assemblies and as a transcriptional co-activator[1][2][3]. LPP is critical for dynamic cytoskeletal remodeling, cell adhesion, motility, and migration through interactions with actin-binding proteins, and is modulated by phosphorylation and protein–protein interactions[1][3]. The gene is frequently involved in chromosomal translocations resulting in fusion proteins that contribute to tumorigenesis—particularly benign soft tissue tumors such as lipomas, but also implicated in a broad range of immune, neoplastic, and autoimmune diseases through genetic variants or altered expression[1][2][7]. No direct approved drugs or clinical inhibitors specifically target LPP, but its expression and mutations serve as important biomarkers for disease susceptibility, tumor subtyping, and evaluation of certain pathologies[1][3].
