LIM domain-containing protein 1 (LIMD1) is a tumor suppressor encoded on chromosome 3p21.3 that belongs to the Zyxin family of LIM-domain proteins. It acts as a multifunctional scaffold involved in transcriptional regulation, cell cycle control, and protein complex assembly. LIMD1 exerts key tumor-suppressive effects by binding to and enhancing the function of retinoblastoma protein (pRB), repressing E2F1-driven gene expression, and inhibiting cell proliferation. It also participates in the microRNA-induced silencing complex (miRISC) and facilitates the degradation of HIF-1α, thus modulating hypoxic response. Downregulation or loss of LIMD1 expression is observed in various solid tumors, including lung, breast, and head and neck cancers. While not an enzyme, transporter, or receptor, LIMD1 is an important mediator of protein-protein interactions central to oncogenic processes and presents a proposed synthetic lethal vulnerability for targeted therapy in LIMD1-deficient tumors.
