LIM domain-containing protein 2 is a small, LIM-only domain protein that shuttles between the cytoplasm and nucleus and binds partners in both compartments. It contains a classic LIM domain architecture (a double zinc finger structure that facilitates protein-protein interactions) and directly interacts with integrin-linked kinase (ILK), a central mediator of cell adhesion and migration. Overexpression of LIMD2 is consistently correlated with increased tumor cell motility, invasion, and higher metastatic grade in cancers such as thyroid, lung, bladder, melanoma, and breast tumors. LIMD2's function is closely tied to remodeling the actin cytoskeleton and facilitating metastatic spread, making it a promising biomarker for aggressive cancers and a potential target for the development of anti-metastatic therapies.
