LIM domain only protein 3 (LMO3) is a cysteine-rich transcriptional co-factor of the rhombotin family, predominantly expressed in the brain, and functions as a context-dependent transcriptional regulator. LMO3 is implicated in the pathogenesis of several tumors, most notably as an oncogene in neuroblastoma, lung adenocarcinoma, and glioma, where its overexpression correlates with poor prognosis. LMO3 forms complexes with neuronal transcription factors such as HEN2, promotes transcription of cell proliferation regulators, and interacts with the tumor suppressor p53 as a co-repressor, modulating p53-dependent transcriptional responses. While essential for transcriptional regulation processes in neuronal tissue, compensatory mechanisms may exist among LIM-only protein family members; there are currently no approved drugs that specifically target LMO3.
