Overview

LIM homeobox 9 is a developmentally expressed transcription factor containing a homeodomain and two LIM domains, enabling both DNA binding and protein-protein interactions. It plays essential roles in early gonadal development—acting with WT1 to activate Sf1, which is necessary for proper development of sexual organs. In the nervous system, LHX9 controls neuronal subtype specification and axon guidance, ensuring correct wiring of spinal pathways and defining neuronal identity in various brain regions. It is crucial for the development of the pineal gland and for maintaining normal neuroanatomy. LHX9 is highly conserved across evolution and participates in a network of transcriptional regulators coordinating organogenesis. In certain cancers, such as pediatric malignant astrocytoma and hepatocellular carcinoma, LHX9 is often epigenetically silenced, consistent with its proposed tumor suppressor function; its silencing is associated with increased cell migration and invasiveness
