Overview

LIM homeobox protein 2 (LHX2) is a transcription factor that belongs to the LIM-homeodomain protein family, characterized by the presence of a homeobox DNA-binding domain and two LIM domains which mediate protein–protein interactions. LHX2 regulates gene expression by functioning as a transcriptional activator or repressor depending on cellular context. It plays crucial roles in neural induction, morphogenesis, differentiation of lymphoid and neural cells, eye and hair development, olfactory sensory neuron specification, liver differentiation, and hematopoiesis. In bone, LHX2 acts as a negative regulator of osteoclast differentiation and is involved in bone remodeling by interacting with signaling molecules such as c-Fos. LHX2 dysfunction or altered expression is associated with developmental disorders and may contribute to pathological bone loss, but it is not currently a direct drug target and no approved pharmacological modulators are available[1][2][3][6].
