Overview

LIM homeobox protein 5 (Lhx5) is a transcription factor characterized by containing two LIM domains and a central homeodomain, allowing it to bind DNA and regulate the expression of downstream genes crucial for development[3][4]. It is most highly expressed in early stages of brain development, particularly in the hypothalamic mamillary body, hippocampus, and hindbrain. Loss of Lhx5 results in severe neurodevelopmental defects in mice, including lack of mamillary body formation and deficits in neuron migration, memory, and motor function[3]. While other LIM homeobox family members are associated with cancer progression and biomarker potential, Lhx5 itself has not yet been directly implicated as a therapeutic target or marker in humans[1].
