Overview

LIM zinc finger domain containing 2 (LIMS2) is an adapter protein found in the cytoplasm where it forms part of a complex that links beta-integrins to the actin cytoskeleton and bridges these complexes to cell surface receptor tyrosine kinases and growth factor receptors[1][2][6][7][8]. LIMS2 is important for modulating cell shape, cell spreading, and migration by organizing protein-protein interactions at focal adhesions, which are cellular structures that mediate connection between the cell and extracellular matrix. The protein contains five LIM domains, each forming two zinc fingers, which mediate these interactions. Pathogenic mutations in LIMS2 are associated with a rare form of autosomal recessive muscular dystrophy with cardiomyopathy and triangular tongue, and emerging studies support a potential role in cancer biology as a suppressor of colon cancer cell migration[1][2][6][9]. No approved drugs or clinical biomarkers are currently described for LIMS2 as a direct therapeutic target.
