LIN37 is a conserved protein that serves as an essential subunit of the MuvB core and specifically the DREAM repressor complex, which regulates cell cycle-dependent gene expression[1][2][3][9]. In quiescent cells, LIN37 binds to LIN9 and RBAP48 via its CRAW domain and partners with other MuvB subunits (LIN9, LIN52, LIN54, RBAP48) to assemble the DREAM complex[1]. This complex represses a broad range of G1/S and G2/M cell cycle genes by recruiting chromatin remodeling proteins, stabilizing nucleosomes downstream of cell cycle promoters, and preventing the activation of cell division and DNA repair transcriptional programs[7][10]. Loss of LIN37 abolishes DREAM-mediated gene repression, leading to uncontrolled cell proliferation and failed quiescence, but does not affect the transcriptional activator forms of MuvB (MMB and FOXM1-MuvB)[3]. LIN37 is not known to be mutated as a cause of disease but its essential role in DREAM highlights its relevance in models of cancer and growth control[10][12][13]. There are no reported drugs or biomarkers for direct clinical targeting or monitoring of LIN37. LIN37 does not fit into standard target categories such as receptor, enzyme, transporter, or ion channel; it is best classified as a **transcriptional coregulator protein complex constituent**.
