The LINE-1 retrotransposon endonuclease (L1-EN), specifically the endonuclease domain of ORF2p, is an enzyme responsible for initiating the retrotransposition of LINE-1 elements. It catalyzes a site-specific single-strand nick in genomic DNA, providing a free 3' hydroxyl group that serves as a primer for reverse transcription.
