Overview

LINE-1 type transposase domain-containing protein 1 (L1TD1) is a domesticated protein derived from the LINE-1 (L1) retrotransposon, functioning as an RNA-binding protein. L1TD1 is mainly expressed in early embryonic cells and select cancer types, where its expression is regulated epigenetically (suppressed by DNA methylation, induced by hypomethylation). Functionally, L1TD1 associates with LINE-1 ribonucleoprotein complexes and promotes L1 retrotransposition in human cells. This protein shows affinity for both LINE-1 and SINE transposon transcripts and is involved in modulating cell cycle regulators, apoptosis, and cell viability, with effects that may vary by cancer type. L1TD1 is not a classical drug target (e.g., receptor, enzyme, transporter) but is under investigation as a potential cancer biomarker due to its role in cell proliferation, apoptosis, and its restricted expression pattern[1][2].
