LINGO1 antisense RNA 2 (LINGO1-AS2) is a long non-coding RNA transcribed from the LINGO1 locus in the antisense direction relative to the LINGO1 protein-coding gene. Like other antisense lncRNAs, LINGO1-AS2 does not code for a protein but is thought to play a regulatory role over LINGO1 gene expression, primarily through mechanisms possible in cis (acting near its own locus). Antisense lncRNAs can regulate their sense gene partners by affecting transcription, promoter/enhancer activity, chromatin structure, or by direct RNA-mediated action. In general, antisense RNAs may serve either oncogenic or tumor-suppressive functions and are increasingly being viewed as therapeutic targets or agents, although there is currently no direct evidence or clinical application for LINGO1-AS2 itself. More broadly, LINGO1 protein itself is implicated as a central regulator of neuronal survival, myelination, and axonal regeneration in the central nervous system, but LINGO1-AS2's exact biological and pathological roles remain to be clarified.
Antisense lncRNAs in general may regulate neighboring gene expression, sometimes by modulating chromatin state, interfering with transcription, or RNA-RNA interactions, but no specific mechanism of action for drugs targeting LINGO1-AS2 is known.
