The linoleic acid pathway encompasses the metabolic conversion of linoleic acid – an essential omega-6 fatty acid – into longer-chain polyunsaturated fats and bioactive lipid mediators, such as gamma-linolenic acid, dihomo-gamma-linolenic acid, and arachidonic acid. Enzymes including desaturases, elongases, cyclooxygenases, lipoxygenases, and cytochrome P450s drive these reactions. The generated metabolites are crucial for the synthesis of signaling molecules (particularly eicosanoids) which regulate inflammation, immune responses, cell growth, and cardiovascular function. While essential in moderate amounts for health (notably skin, neuronal, and immune function), excess intake and oxidative modification of linoleic acid and its derivatives have been associated with various chronic diseases including heart disease, cancer, chronic inflammation, and neurodegenerative disorders. Epigenetic modifications in genes of the pathway may influence psychological resilience and the severity of disorders like PTSD.
