Lipase member I (LIPI) is an enzyme within the phospholipase A1 family, sometimes designated as “membrane-associated phosphatidic acid-selective phospholipase A1-beta.” It catalyzes the hydrolysis of phospholipids, releasing fatty acids from the sn-1 position of phospholipids. LIPI is also identified as a cancer/testis antigen (CT17), meaning it is aberrantly expressed in some tumors with restricted normal tissue distribution (primarily testis), making it a potential immuno-oncology target. As a phospholipase, LIPI participates in the breakdown of phospholipids, lipid remodeling, and cell signaling. Aberrant expression or function has been linked to cancer and may also be implicated in lipid metabolic disorders. The structure includes conserved motifs typical of phospholipases and domains associated with membrane binding and catalytic activity. Therapeutic research is mainly in early stages, including exploration as a cancer vaccine or immune target; no known approved drugs selectively inhibit or modulate LIPI[2][3].
