Overview

LIPE-AS1 is a **long non-coding RNA (lncRNA)** encoded on the plus strand of human chromosome 19, transcribed antisense to the gene LIPE (encoding hormone-sensitive lipase), with conservation of gene structure and adipose tissue expression across human and mouse. LIPE-AS1 is highly expressed in subcutaneous and visceral adipose tissue, adrenal gland, testis, and mammary gland, correlating with the tissue distribution of LIPE itself. Functional studies show that knockdown of LIPE-AS1 variants can suppress adipogenesis and promote apoptosis in pre-adipocyte cell lines by downregulating adipogenic transcription factors (such as Pparg, Cebpa) and upregulating apoptosis markers. This suggests a regulatory role in protecting against stress during fat cell differentiation[2]. There is currently no evidence for its role as a therapeutic target, no reported drug ligands, nor established utility as a clinical biomarker.
