Overview

Lipid bilayer membrane microdomains, commonly known as lipid rafts, are small, dynamic, and highly organized areas within the cell membrane enriched in cholesterol and sphingolipids. These domains act as specialized platforms that concentrate specific proteins, thereby facilitating efficient signal transduction and membrane trafficking (Simons & Ikonen, 1997; Lingwood & Simons, 2010). They play a crucial role in various biological processes, including endocytosis, cell-cell communication, and the regulation of the cytoskeleton. In many diseases, the structural integrity or composition of these microdomains is altered; for instance, they can facilitate the entry of pathogens like HIV or the aggregation of toxic proteins in Alzheimer's disease (Michel & Bakovic, 2007). In oncology, lipid rafts often sequester pro-survival signaling molecules, making them attractive targets for anti-cancer therapies (Mollinedo & Gajate, 2015). Therapeutic strategies targeting these domains, known as membrane lipid therapy, utilize drugs to modulate membrane fluidity or deplete essential lipid components like cholesterol to disrupt pathological signaling (Escribá et al., 2008).
