Overview

Lipid I is a vital membrane-anchored intermediate in the bacterial peptidoglycan biosynthetic pathway, consisting of a MurNAc-pentapeptide linked to an undecaprenyl lipid carrier via a pyrophosphate bridge (Ling et al., 2015, Nature). This pyrophosphate moiety is a highly conserved structural motif that is essential for the subsequent conversion of Lipid I into Lipid II by the enzyme MurG (Breukink & de Kruijff, 2006, Nature Reviews Drug Discovery). Because of its critical role and accessibility on the inner leaflet of the cytoplasmic membrane, the Lipid I pyrophosphate has emerged as a prime target for resistance-proof antibiotics like teixobactin and clovibactin (Shukla et al., 2023, Cell). These antimicrobial agents bind to the pyrophosphate group through a network of hydrogen bonds, effectively sequestering the precursor and halting cell wall assembly. This mechanism leads to rapid bacterial cell death and lysis, particularly in Gram-positive pathogens (Hover et al., 2018, Nature Microbiology). Since the target is a non-proteinaceous, highly conserved metabolic intermediate, the development of resistance through target modification is significantly hindered compared to traditional protein-targeting antibiotics.
