Lipin 1 is a multifunctional protein that functions as a phosphatidate phosphatase, converting phosphatidic acid to diacylglycerol, and as a transcriptional coactivator regulating genes involved in lipid metabolism and energy balance. It plays a critical role in glycerolipid synthesis, adipocyte differentiation, and the regulation of fatty acid oxidation. Mutations in LPIN1 are associated with recurrent rhabdomyolysis and a predisposition to statin-induced myopathy. It is also implicated in lipodystrophy syndromes.
