Overview

Lipoma HMGIC fusion partner-like 3 (LHFPL3) is a tetraspan transmembrane protein belonging to the lipoma HMGIC fusion partner (LHFP) gene family, a subset of the larger tetraspan superfamily[1][2][4]. LHFPL3 is primarily recognized for its emerging roles in the nervous system, where it may regulate synaptic processes and contribute to neurodevelopment[1]. It is highly expressed in certain cancers, particularly malignant glioma, and may be involved in tumor pathogenesis via modulation by regulatory microRNAs, potentially serving as a biomarker for glioblastoma subtypes[1]. Deletions at the LHFPL3 locus have been linked to benign uterine leiomyomas and overgrowth syndromes, suggesting a function in cell growth and cell cycle regulation[1]. Although LHFPL3 has been implicated in cancer and neurodevelopmental processes, there is currently no evidence that it serves as a therapeutic target or that any drugs directly act on it[2][4]. Note: LHFPL3 is sometimes confused with LHFPL4 (another alias/previous symbol), but LHFPL3 is the canonical, current designation[4]. No drugs or mechanisms of action are known for this protein, and it is not a validated therapeutic target at this time. However, its involvement in synaptic regulation and tumor biology is under investigation for future roles as a biomarker or potential target[1][4].
