Lipopolysaccharide export system protein LptD (LptD) is a large β-barrel outer membrane protein essential for the final step of transporting lipopolysaccharide (LPS) from the periplasm to the external surface of Gram-negative bacteria[3][5][2]. LptD forms a highly stable 1:1 complex with the lipoprotein LptE and is structurally composed of an N-terminal periplasmic β-jellyroll domain and a C-terminal transmembrane β-barrel with 26 antiparallel strands[3][2][5]. This complex acts as a channel, facilitating the unidirectional insertion of LPS into the outer membrane, which is critical for bacterial viability and membrane integrity[3][5][2]. LptD is part of the Lpt pathway (LptABCDEFG), all of which are essential for LPS transport and thus are potential targets for antibiotics against multidrug-resistant Gram-negative pathogens[3][5].
